The Inhibition of Human Placental Diamine Oxidase

چکیده
1. The oxidation of p-dimethylaminomethylbenzylamine by purified placental diamine oxidase was followed by measuring the change in E250 caused by the production of p-dimethylaminomethylbenzaldehyde. 2. The inhibition of this reaction by substrate analogues such as isothiouronium, guanidinium, dimethylsulphonium and trimethylammonium compounds was extensively studied. 3. The type and degree of inhibition by monoand bis-onium compounds is described, and a theory is developed to explain the type of inhibition produced. 183

The amine oxidases of human placenta and pregnancy plasma.
1. The purification of monoamine oxidase and diamine oxidase from normal human term placental tissue is described. 2. The properties of these enzymes are reported and compared with the properties of unpurified human pregnancy plasma. 3. This comparison shows that the amine oxidase of pregnancy plasma has properties corresponding to purified placental diamine oxidase, suggesting a placental orig...
متن کاملDiamine oxidase is the amiloride-binding protein and is inhibited by amiloride analogues.
Diamine oxidase (histaminase), an enzyme that oxidatively deaminates putrescine and histamine, was purified from human placenta and from pig kidney. Both NH2-terminal sequences are highly homologous to the human kidney amiloride-binding protein, previously thought to be a component of the amiloride-sensitive Na+ channel. Monoclonal antibodies raised against the pig kidney amiloride-binding prot...
متن کاملThe purification and preliminary investigation of fumarase, peroxidase, diamine oxidase and adenosine deaminase from human seminal plasma [proceedings].
The biochemistry of human semen is poorly understood, and despite initial studies by Mann (1964) little is known about the metabolism of spermatozoa1 cells, and even less about the seminal plasma. We have attempted the simultaneous purification of fumarase, peroxidase, diamine oxidase and adenosine deaminase from human seminal plasma, and compared the properties of these enzymes with those from...
متن کاملNutrients regulate diamine oxidase release from intestinal mucosa.
Diamine oxidase is continuously released from the intestinal mucosa and carried to the circulation by the lymphatics. The effect of nutrients on this release was examined. Rats were prepared with duodenal and intestinal lymph cannulas. Test mixtures of lipid emulsions containing triolein, oleic acid, or tricaprylin and solutions of carbohydrate and protein were infused into the duodenum. The en...
متن کاملDiamine oxidase and cadaverine metabolism.
We have recently presented evidence that diamine oxidase is rcsponsiblc for a portion of the metabolism of histamine in several species (1, 2). This evidence is largely based on the alteration of histamine metabolism produced by compounds which are known to inhibit diamine oxidase in vitro. The inhibitors used were isonicotinylhydrazine (3) and aminoguanidine. The latter is reported to be the m...
